Sampling from high viscosity and slurry cultures

Features

  • Sample volume adjustable 2-10 mL per pull; several pulls per sampling cycle possible
  • Sampling interval c. 10 min, but depends on the length of the analysis
  • Sample at the analyser 2 min after sampling
  • 1st filtration stage adjustable during sampling eliminates >500 µm particles
  • 2nd filtration stage in the piston eliminates >200 µm particles
  • Sterilization and cleaning with 2 bar steam
  • Drying with 2 bar air or nitrogen
  • Connection to reactor or pipe into 25 mm Ingold-style port
  • Connection to Filtration Module: Sample sent with overpressure via 1/8” Teflon tube

Filtration Module

Filtration Module

Filtration Module performs the final filtration of the sample delivering a particle-free sample.

Features

  • 3rd filtration stage in the Filtration module eliminates remaining >0.2 µm particles in a circulated cross-flow filtration using round 45 mm filter membranes
  • Cleaning with steam condensate
  • Drying with 2 bar air or nitrogen
  • Connection to Dilution Module: Sample sent with overpressure via 1/8” Teflon tube

Dilution Module

Dilution Module

Dilution Module performs automatically up to 20-fold dilution of the particle-free sample. Dilution Module can also be used to prepare the dilution series of standards for automated calibration of the on-line analyser.

Online HPLC

Online HPLC

Online HPLC enables automated analyses of the samples. One HPLC pump is used for isocratic separation methods and an additional pump can be connected for gradient methods. Refractive index (RI), Ultraviolet (UV) or even electrospray mass spectrometer (MS) can be used as detectors.

  • Injection valve with 10-100 µL injection loop (full loop injection)
  • Eluent degasser
  • One or two HPLC pumps with 0.1-5 mL flow rate
  • Column oven; temperature range +30..+70 °C
  • RI and/or UV detector
  • As an example Aminex HPX-87H column enables analysis of glucose, xylose, sucrose, xylitol, glycerol, acetate, lactate, ethanol, butanol, succinate in 18 minutes using 5 mM sulphuric acid as eluent
